Bacterial-Like Nonribosomal Peptide Synthetases Produce Cyclopeptides in the Zygomycetous Fungus Mortierella alpina

چکیده

Fungal natural compounds are industrially produced, with application in antibiotic treatment, cancer medications, and crop plant protection. Traditionally, higher fungi have been intensively investigated concerning their metabolic potential, but reidentification of already known is frequently observed. Hence, alternative strategies to acquire novel bioactive molecules required. We present the genus Mortierella as representative early-diverging an underestimated resource products. alpina produces two families cyclopeptides, designated malpicyclins malpibaldins, respectively, via pentamodular nonribosomal peptide synthetases (NRPSs). These enzymes much more closely related bacterial than other fungal NRPSs, suggesting a origin these NRPS genes . Both were biochemically characterized involved as-yet-unknown biosynthetic pathways products basal fungi. this report establishes prolific compound producers sheds light on capacity.

Genome Characterization of the Oleaginous Fungus Mortierella alpina

Mortierella alpina is an oleaginous fungus which can produce lipids accounting for up to 50% of its dry weight in the form of triacylglycerols. It is used commercially for the production of arachidonic acid. Using a combination of high throughput sequencing and lipid profiling, we have assembled the M. alpina genome, mapped its lipogenesis pathway and determined its major lipid species. The 38....

Ice nucleation activity in the widespread soil fungus Mortierella alpina

Biological residues in soil dust are a potentially strong source of atmospheric ice nuclei (IN). So far, however, the abundance, diversity, sources, seasonality, and role of biological – in particular, fungal – IN in soil dust have not been characterized. By analysis of the culturable fungi in topsoils, from a range of different land use and ecosystem types in southeast Wyoming, we found ice-nu...

Dissecting and exploiting nonribosomal peptide synthetases.

A large number of therapeutically useful cyclic and linear peptides of bacteria or fungal origin are synthesized via a template-directed, nucleic-acid-independent nonribosomal mechanism. This process is carried out by mega-enzymes called nonribosomal peptide synthetases (NRPSs). NRPSs contain repeated coordinated groups of active sites called modules, and each module is composed of several doma...

Phylogenetic analysis of condensation domains in the nonribosomal peptide synthetases.

Condensation (C) domains in the nonribosomal peptide synthetases are capable of catalyzing peptide bond formation between two consecutively bound various amino acids. C-domains coincide in frequency with the number of peptide bonds in the product peptide. In this study, a phylogenetic approach was used to investigate structural diversity of bacterial C-domains. Phylogenetic trees show that the ...

Polyketide synthases and nonribosomal peptide synthetases: the emerging view from bacterial genomics.

A total of 223 complete bacterial genomes are analyzed, with 281 citations, for the presence of genes encoding modular polyketide synthases (PKS) and nonribosomal peptide synthetases (NRPS). We report on the distribution of these systems in different bacterial taxa and, whenever known, the metabolites they synthesize. We also highlight, in the different bacterial lineages, the PKS and NRPS gene...
